A map of the heart and a sequence of diagrams help locate the different parts and explain the pumping cycle. Four hollow chambers fortified with blood vessels make up this muscular organ. Two thin-walled ante-rooms, the left atrium and the right atrium, serve as holding tanks for blood entering the heart;thick walled ventricles, one under each atrium, perform the serious labor of pumping blood. An impermeable wall divides the right and left sides of the heart.
The heart fills and empties in a second long rhythmic cycle of contraction and relaxation, a heartbeat. Dark oxygen-spent blood(blue) from the body converges into two great veins, the superior and inferior venae cavae, en route to the right atrium, while bright red oxygenated blood moves towards the left atrium from the lungs via the pulmonary veins. As ventricles relax, a phase called diastole, the atria fill with blood and force open two valves through which the blood drains into the left and right ventricles. In the pumping, or systole, phase, the ventricle contract, closing off the two lower valves. The right ventricle forces the oxygen spent blood into the pulmonary artery;the left ventricle pumps oxygenated blood into the aorta and on into the body's tissues.
North America has one of the highest heart disease rates in the world. Since 1940 cardiovascular disease(CVD) has been the leading cause of mortality and morbidity in the region. Abstract: The role of herbal drugs in preventing myocardial infraction and prevention of cardiovascular diseases is increasing with the inability of conventional therapeutic agents to control or reverse disease. Many herbs have been used in various traditional systems of medicine for cardiac problems. One such polyherbal formula is AV CIRCULO. The main objective of the study was to evaluate the cardioprotective activity of AV CIRCULO in isoproterenol induced myocardial necrosis in rats.

Methods: Acute toxicity and behavioral studies were carried out in albino mice by oral route at various doses of AV CIRCULO. A model for cardiac necrosis was established by administering different doses of isoproterenol, subcutaneously for 2 consecutive days. AV CIRCULO was administered orally daily prophyllactically to rats for a period of 20 days(sub acute treatment) and also for a period of 45 days( chronic treatment) at different doses. During the last two days of AV CIRCULO treatment, isoproterenol was administered subcutaneously for 2 consecutive days. The animals were sacrificed after 24 hours of the second dose of isoproterenol. Biochemical parameters like changes in ratio of heart weight to body weight was studied. Serum enzyme levels of creatinine phosphokinase ( CPK), lactate dehydrogenase(LDH), glutamic oxaloacetic transaminase(GOT), serum lipid levels ( Triglycerides and total cholesterol), total proteins and DNA content of heart were also studied. Quantitative measurement of lipid peroxidation was also determined by measuring the concentration of thiobarbituric acid reactive substances (TBARS) in heart homogenate. Histopathology of heart section was examined to assess the architectural changes in cardiac muscle.

Results & Discussions: Isoproterenol treatment had induced myocardial necrosis in rats. The pretreatment with AV CIRCULO both at subacute and Chronic period significantly prevented the increase in serum enzyme levels of LDH,CPK & SGOT due to isoproterenol treatment. However the treatment of 45 days with AV CIRCULO showed much better results than 20 days of pre treatment.The present findings of the biological and biochemical parameters suggest a significant cardioprotective activity of AV CIRCULO against isoproterenol induced myocardial necrosis.


Terminalia arjuna bark
This herb has been conclusively proven to have heart protective and blood pressure lowering activity. Studies have shown that extracts relieved heart pain and lowered blood pressure. It has also been shown to improve irregular palpitations. It increases the force of contraction and assists the nutrient supply to the heart, resulting in the heart muscle gaining energy and the number of heartbeats is reduced.

Crategus oxycantha
The fruits are cardiotonic, coronary vasodilator. It is reputed to dissolve deposits in thickened and sclerotic arteries. Various clinical studies reveal increased cardiac performance and output: decrease in peripheral vascular resistance; decrease in pulmonary arterial and capillary pressures; reductions in blood pressure at rest and during exercise; and improved metabolic parameters.This herb improves circulation and nutrition to the heart by dilating the coronary blood vessels. Coleus forskholii (Forskholin 4%)
Detailed pharmacological studies established that forskohlin lowered normal or elevated blood pressure in different animal species through a vasodilatory effect and that it had a positive intopic action on the heart muscle . The antihypertensive effects of forskohlin were attributed to a decrease in peripheral resistance by action of the drug on arteriolar smooth muscle. The positive inotropic effects of forskohlin in the cardiac muscle were shown to be initiated by stimulation of the myocardium cell membrane adenylate cyclase, followed by a rise in cyclic AMP, an activation of membrane-bound protein kinases, a lowering of the membrane bound Na+, K+ -ATPase activity and activation of the slow calcium gate, and an activation of cytopalsmic protein kinases. Coleus Forskohlii, shows that the plant kingdom offers excellent prospects in the search for further potent cardiac agents.

Boerhaavia diffusa
The cardiovascular action of this herb has been clinically studied. Due to its diuretic activity it is specific for heart conditions with fluid retention.

Withania somnifera
Withania somnifera has been variously described as the most important medicinal plant in India. It has been used in most preperations due to it's confimed immunomodulatory action. This herb has been the focus of hundreds of clinical studies and is one of the most exhaustively researched herbs. Pharmacology of Forskolin – Cardiology

The anti-hypertensive effects are attributed to a decrease in pre and after loads andimproved left ventricular function as well as decreased peripheral resistance by a directaction on relaxing the arterial smooth muscle. Forskolin also has a positive inotropiceffect on heart muscle. Both the positive inotropic action and the vascular action wereshown to be initiated by the stimulation of the membrane bound enzyme adenylatecyclase resulting in raised intracellular cAMP levels, an activation of protein synthesis, alowering of the membrane bound Na+, K+-ATPase activity and activation of the slowcalcium gate. Stimulation of cAMP levels increases the contractility of the myocardium.Adenylate cyclase is an enzyme-system-complex, with various activating catalytic unitsand sub-units embedded in the membrane bi-layers of numerous tissues including themyocardium, bronchioles, arteries, kidneys etc. Adenylate cyclase has 2 polar sub-units –the catalytic C unit and the nucleotide regulatory sub-unit N. Forskolin directly stimulatesthe C sub-unit, resulting in stimulation of cAMP, which in turns activates and elicits acascade of enzymatic functions.Beta-adrenergic agonists are potent pharmacological cardiac agents and are usedtherapeutically in the treatment of heart failure. However, such compounds have a majordisadvantage that limits their long term use. Continuous exposure to beta-agonistsrenders the myocardium sub-sensitive by reducing beta-receptor density and/or receptorcyclasecoupling. Both of these sub-sensitivity phenomenon are apparently regulatedthrough the N sub-unit of Adenylate Cyclase. Forskolin, however, acts primarily via the Csub-unit and does not demonstrate the sub-sensitivity phenomenon or beta-receptor downregulation. Indeed Forskolin exhibits positive inotropic and chronotropic characteristics, asseen in guinea pig, sheep, rat, pig, rabbit and human studies.Forskolin also been shown to inhibit platelet aggregation and produces deaggregation ofhuman aggregated platelets.NOTE: The Forskolin content in AV CIRCULO is optimally standardized by HPLC. TheFull spectrum extract containing 4% Forskolin content is the maximal amount extractable withoutcompromising the quality and exclusion of other beneficial compounds. Higher levels ofForskolin results in a less effective product. This is confirmed by the studies of Hoechst AG, who conducted the original research.
